Believed because innate immune cells lack the antigen receptors that give T and B cells their memory. Corrected in 2009–2011 by discovering NK cells can be epigenetically ‘primed’ by a past exposure — explaining why the BCG vaccine gives broad, non-specific protection beyond just tuberculosis.
The intuitive logic seemed obvious: more protection must be better. In reality, an overactive immune system causes lupus, severe allergy, and cytokine storms — a 1990s high-dose ‘immune-boosting’ cancer therapy caused dangerous toxicity for exactly this reason.
Long assumed because the brain sits behind a protective barrier, seemingly walled off from immunity. The 2015 discovery of lymphatic vessels draining the brain’s own lining proved it was never truly isolated — launching the field connecting chronic stress directly to immune decline.
It shrinks with age and was long dismissed as vestigial — doctors even used it as an X-ray target in children for decades, unknowingly raising their future thyroid cancer risk. A single 1961 mouse experiment proved it’s essential to the entire T cell system.
The original theory held that self-reactive immune cells get permanently deleted early on, and that’s the end of it. A 2003 discovery showed a dedicated cell type keeps actively enforcing tolerance every day — proven by a rare genetic condition where losing this cell type causes severe autoimmune disease across multiple organs.
Today’s assumptions that might not survive: many specific gut-microbiome disease claims, treating a single molecule as the sole ‘driver’ of a disease, and assuming immune aging can never be reversed.
